Pathological changes leading to illness have been confirmed for Alzheimer’s (amyloid plaques) and Parkinson’s disease (loss of dopaminergic transmission) and are the foundation for studies on the molecular biology of these disorders. For other neuropsychiatric disorders, in particular for schizophrenic and affective disorders, molecular causes appear to be more complex and therefore remain hypothetical, despite decades of research. Changes in a number of neuronal pathways and structures have been reported to be associated with these disorders and are currently under extensive investigation. The present volume reviews recent knowledge with emphasis on ongoing research findings. Current hypotheses based on these findings are described and discussed.

Book excerpt: The development of more effective treatments for neuropsychiatric disorders requires scientific progress on a broad front. Animal models have a vital role to play in advancing the field. When deployed in conjunction with detailed study of these diseases in man they bring the power to make controlled experimental interventions which allow the functional consequences of genetic variations and polymorphisms to be understood in terms of their cellular, systems and behavioural effects. Further, they provide a means by which complex cognitive and behavioural phenomena may be dissected and understood. Finally, they provide a bridge to understanding the effects of drugs on the functioning of the central nervous system, thereby improving our understanding of the actions of those drugs in man.

Available in PDF, EPUB and Kindle. Book excerpt: Neuropsychiatric Disorders and Epigenetics, Second Edition is a comprehensive reference on the epigenetic basis of common neuropsychiatric disorders. The volume is organized into chapters covering individual neuropsychiatric disorders, from addiction to anxiety and autism spectrum disorders, and is contributed by leading experts in their respective fields. The epigenetic aspects of each disorder are discussed, in the context of the full range of associated epigenetic mechanisms, including DNA modification, histone post-translational modification, chromatin organization, and non-coding RNA. A particular emphasis is placed on potential epigenetic interventions, when the effects of environmental stimuli on epigenetic states is particularly relevant to disease. This new edition has been fully updated to reflect recent research advances enabled by genomic technologies, as well as therapeutic interventions for previously unmanageable disorders. Several new chapters have been added on disorders or approaches not considered in the earlier edition, including epigenetics and anxiety disorders, epigenetics and neuroimaging in neuropsychiatric disorders, genome-wide approaches to epigenetic research, and the epigenetics of spinal muscular atrophy. By helping to define epigenetics as a key player in neuropsychiatric disorders, this volume empowers new research, clinical translation, and pharmacological advances, and highlights promising directions for ongoing investigation. Analyzes the effects of environmental stimuli on epigenetic states that correlate with neuropsychiatric disease induction Reviews the epigenetic basis for common neuropsychiatric disorders, thereby guiding translational therapies for clinicians and mechanistic studies for scientists Features extensive use of diagrams, illustrations, tables, and graphical abstracts for each section to reinforce understanding Includes chapter contributions from leading global experts

Book excerpt: The Molecular Biology of Neurological Disease reviews advances that have been made in understanding the molecular mechanisms of neurological disorders as well as immediate and future applications of molecular biological techniques to clinical practice. This book explores the molecular genetics of neurological disease such as muscular dystrophy, Joseph disease, and Huntington's disease, along with the mitochondrial genes implicated in such conditions. This text is comprised of 18 chapters and begins by introducing the reader to the basic principles and methods of molecular genetic techniques used in the diagnosis of neurological disease. Attention then turns to several aspects of genetic expression in the brain, including the extent to which the genome is expressed in the brain. The next chapter focuses on the visualization of polyadenylated messenger RNAs in individual cells in mammalian brain using in situ hybridization techniques, combined with immunohistochemical localization of specific proteins and neuropeptides implicated in diseases such as Alzheimer dementia. This book also discusses the molecular biology of chemical synaptic neurotransmission; proteins involved in the regulation of nervous system development; and gene expression in skeletal muscle. This text then concludes with a summary of the ""neurological gene map"" as it stands in the latter part of 1987. This book is intended for physicians who grapple with the problems of neurological disorders on a daily basis, including neurologists, neurologists in training, and those in related fields such as neurosurgery, internal medicine, psychiatry, and rehabilitation medicine.

Available in PDF, EPUB and Kindle. Book excerpt: Our understanding of the neurobiological basis of psychiatric disease has accelerated in the past five years. The fourth edition of Neurobiology of Mental Illness has been completely revamped given these advances and discoveries on the neurobiologic foundations of psychiatry. Like its predecessors the book begins with an overview of the basic science. The emerging technologies in Section 2 have been extensively redone to match the progress in the field including new chapters on the applications of stem cells, optogenetics, and image guided stimulation to our understanding and treatment of psychiatric disorders. Sections' 3 through 8 pertain to the major psychiatric syndromes-the psychoses, mood disorders, anxiety disorders, substance use disorders, dementias, and disorders of childhood-onset. Each of these sections includes our knowledge of their etiology, pathophysiology, and treatment. The final section discusses special topic areas including the neurobiology of sleep, resilience, social attachment, aggression, personality disorders and eating disorders. In all, there are 32 new chapters in this volume including unique insights on DSM-5, the Research Domain Criteria (RDoC) from NIMH, and a perspective on the continuing challenges of diagnosis given what we know of the brain and the mechanisms pertaining to mental illness. This book provides information from numerous levels of analysis including molecular biology and genetics, cellular physiology, neuroanatomy, neuropharmacology, epidemiology, and behavior. In doing so it translates information from the basic laboratory to the clinical laboratory and finally to clinical treatment. No other book distills the basic science and underpinnings of mental disorders and explains the clinical significance to the scope and breadth of this classic text. The result is an excellent and cutting-edge resource for psychiatric residents, psychiatric researchers and doctoral students in neurochemistry and the neurosciences.

Available in PDF, EPUB and Kindle. Book excerpt: This new volume in the Handbook of Clinical Neurology presents a comprehensive review of the fundamental science and clinical treatment of psychiatric disorders. Advances in neuroscience have allowed for dramatic advances in the understanding of psychiatric disorders and treatment. Brain disorders, such as depression and schizophrenia, are the leading cause of disability worldwide. It is estimated that over 25% of the adult population in North America are diagnosed yearly with at least one mental disorder and similar results hold for Europe. Now that neurology and psychiatry agree that all mental disorders are in fact, "brain diseases," this volume provides a foundational introduction to the science defining these disorders and details best practices for psychiatric treatment. Provides a comprehensive review of the scientific foundations of psychiatric disorders and psychiatric treatment Includes detailed results from genetics, molecular biology, brain imaging, and neuropathological, immunological, epidemiological, metabolic, therapeutic and historical aspects of the major psychiatric disorders A "must have" reference and resource for neuroscientists, neurologists, psychiatrists, and clinical psychologists as well as all research scientists investigating disorders of the brain
