Experimental methods have been devised to meas ure the translational and rotational mobility of lipids and proteins, thereby furnishing a quantitative basis for the concept of membrane fluidity. Likewise, the asymmetry of bi layer membranes as evidenced by the asymmetric insertion of proteins and lipids has been put on firm experimental ground. At higher molecular resolution it has been possible to provide a detailed pi2ture of the molecular conformation and dynamics of lipids and, to some extent, even of small peptides embedded in a bilayer matrix. Many of these achieve ments would not have been possible without the application of modem spectroscopic methods. Since these techniques are scattered in a variety of specialized textbooks the present monograph attempts to describe the key spectroscopic methods employed in present-day membrane research at an intermediate level. There is no question that the elusive detailed structure of the biological membrane demands a multiplicity of experi mental approaches and that no single spectroscopic method can cover the full range of physical phenomena encountered in a membrane. Much confusion in the literature has arisen by undue generalizations without considering the frequency range or other limi tations of the methods employed. It is to be hoped that the present monograph with its comprehensive description of most modem spectroscopic techniques, will contribute to- .

Studies in lipid monolayers show the correlation of surface pressure with water activity and its role in peptide and enzyme interactions. The book concludes with a discussion on anhydrobiosis and the effect of water replacement in microdomains and its consequence for cell function. New definitions of lipid/water interphases consider water not only as a structural-making solvent but as a mediator in signalling metabolic activity, modulating protein insertion and enzymatic activity, triggering oscillatory reactions and functioning of membrane bound receptors. Since these effects occur at the molecular level, membrane hydration appears fundamental to understand the behavior of nano systems and confined environments mimicking biological systems. He was a versatile, internationally renowned biochemist, gifted with bound less energy and a great breadth of view. After early work on respiratory enzymes, he developed a lifelong in te rest in complex carbohydrates and, particularly, glycoproteins. He early recognized the important role of the latter substances in membrane function and developed the now widely accepted model for the structure of major erythrocyte membrane glyco pro teins. He expanded this model to include the possible general roles of membrane glycoproteins in specific membrane functions, particularly cell recognition phenomena. Richard Winzler possessed a broad and critical perception of the ftux of biomedical progress. He thus appreciated very early that general recognition of the complexity of membrane-associated phenomena would foster a new, rapidly expanding, interdisciplinary membrane technology. He also recognized the need for means to bridge the communication barriers inevitably genera ted by such an evolution, and this conviction largely stimulated us to write this book. Membrane technology is a vast field and we cannot treat all its aspects. Instead, we concentrate on those areas that, in our view, have evolved most rapidly. In this, we pay primary but not exclusive attention to the plasma membrane, but where relevant, consider other biomem branes and diverse model systems. Moreover, we focus on animal cells, but include information derived from microbial and artificial membranes where germane.
