Book excerpt: Since endothelial progenitor cells (EPCs) were first described in 1997, there has been significant debate surrounding their definition and roles; but also agreement in their potential to develop as biomarkers and cytotherapies. EPCs participate in vascular repair and postnatal angiogenesis by differentiating into endothelial cells or by producing pro–angiogenic growth factors. Various subtypes of EPCs have been studied, such as CD34+VEGFR2+ blood circulating cells, cultured endothelial colony forming cells (ECFCs), and myeloid angiogenic cells (MACs). EPCs have therapeutic potential for revascularisation and vascular repair in ischemic diseases such as myocardial infarction and diabetic vascular complications. In this eBook, we compile evidence to enable their translation including strategies to enhance the number, homing ability to the injury site and function of EPCs.

Available in PDF, EPUB and Kindle. Book excerpt: We are delighted to offer this textbook to the scientific community, entitled Endothelial Progenitor Cells in Health and Diseases, which covers a timely topic in the rapidly evolving discipline of vascular biology. Written primarily for a life science audience and clinicians, the fundamentals of EPCs biology and the latest characterization and definition in health and diseases are introduced, followed by explanations of the most cutting-edge cell therapy methods to cure ischemic diseases. In Section One, endothelial cell progenitor isolation methods and biological characterization were reported. The discovery of this novel endothelial progenitor cells (EPC) concept has overturned the previous dogma which suggested that vasculogenesis could only occur during embryogenesis. In fact, both vasculogenesis and angiogenesis may potentially have a synergistic role in postnatal revascularization. Also, this chapter summarized recent advances in EPC biology such as biological function, origin, definition, and classification. Each EPC culture and isolation method is clearly defined to prevent confusion in EPC biology. Section Two focuses on EPC biological function alteration in cardiovascular diseases (CVD). Several large clinical trials have reported that the number and biological function are strongly associated with major adverse cardiovascular events. The diagnostic and prognostic potential of EPC is crucial in terms of CVD. In Section Three, biological dysfunction mechanisms of EPC and their scarcity in diabetic patients' peripheral blood were clearly described. Preclinical studies have shown that EPC-based therapy is feasible, safe, and efficacious in multiple disease states. Subsequently, this has led to several clinical trials demonstrating the feasibility and safety profile of EPC therapy against cardiovascular ischemic diseases. In Section Four, regenerative medicine pioneers discussed EPCs translation to the clinic and cell transplantation challenges along with their solutions. Personalized stem cell-based therapy approaches employing several clinical biomarkers, disease-related genetic-trait evaluation methods, and advanced analyses with state-of-the-art computational methods such as machine learning-based prediction can increase cell therapies' efficacy and decrease treatment costs. The last chapter in the book describes a therapeutic application of EPC-derived extracellular vesicles to cure CVD.

Book excerpt: Recent scientific breakthroughs, celebrity patient advocates, and conflicting religious beliefs have come together to bring the state of stem cell researchâ€"specifically embryonic stem cell researchâ€"into the political crosshairs. President Bush's watershed policy statement allows federal funding for embryonic stem cell research but only on a limited number of stem cell lines. Millions of Americans could be affected by the continuing political debate among policymakers and the public. Stem Cells and the Future of Regenerative Medicine provides a deeper exploration of the biological, ethical, and funding questions prompted by the therapeutic potential of undifferentiated human cells. In terms accessible to lay readers, the book summarizes what we know about adult and embryonic stem cells and discusses how to go about the transition from mouse studies to research that has therapeutic implications for people. Perhaps most important, Stem Cells and the Future of Regenerative Medicine also provides an overview of the moral and ethical problems that arise from the use of embryonic stem cells. This timely book compares the impact of public and private research funding and discusses approaches to appropriate research oversight. Based on the insights of leading scientists, ethicists, and other authorities, the book offers authoritative recommendations regarding the use of existing stem cell lines versus new lines in research, the important role of the federal government in this field of research, and other fundamental issues.

Available in PDF, EPUB and Kindle. Book excerpt: Since the publication of the first edition of this book in 2010, an explosion of spectacular discoveries in the field of regeneration has compelled the current revisit of the field of Regenerative Nephrology. This second edition features subjects as diverse as age and gender influencing regenerative processes; mechanisms and pathways of premature cell senescence affecting kidney regeneration; the ways intrinsic regenerative processes can become subverted by noxious stressors eventuating in disease progression; novel mechanistic and engineering efforts to recreate functional kidney or its component parts; cell reprogramming and reconditioning as emerging tools of future regenerative efforts; and effects of various biologicals on kidney regeneration. These newer additions to the armamentarium of Regenerative Medicine and Nephrology have become an integral part of the second edition of the book. Cutting-edge investigations are summarized by the constellation of the most experienced contributing authors coming together from around the world under the umbrella of the second edition. A significant expansion of section on induced pluripotent cells and trajectories of their differentiation. This will be followed by mechanisms and modalities of cell reprogramming for therapeutic purposes A new section on tissue engineering of the kidney of interest to nephrologists and urologists An entire section dedicated to causes of regenerative failure with the emphasis on recent discoveries of senescent cells in kidney disease, pathologic effects of senescent cells, advents in senotherapies and rejuvenation therapies A vastly expanded section on pharmacotherapies promoting kidney regeneration, trials of engineered organs, manufacturing in regenerative medicine and smooth transition to the clinical trials, with an update on some ethical issues

Much of this research has been fueled by their potential for use in regenerative medicine applications, which has in turn spurred growing numbers of translational and clinical studies. However, more work is needed if the potential is to be realized for improvement of the lives and well-being of patients with numerous diseases and conditions. This book series 'Cell Biology and Translational Medicine (CBTMED)' as part of Springer Nature’s longstanding and very successful Advances in Experimental Medicine and Biology book series, has the goal to accelerate advances by timely information exchange. Emerging areas of regenerative medicine and translational aspects of stem cells are covered in each volume. Outstanding researchers are recruited to highlight developments and remaining challenges in both the basic research and clinical arenas. This current book is the 17th volume of a continuing series.
