Available in PDF, EPUB and Kindle. Book excerpt: Peptide nucleic acids (PNAs) have now existed for slightly more than ten years, with the interest in and applications of this pseudopeptide DNA mimic steadily increasing during the entire period. PNAs have rapidly attracted the attention of scientists from a diversity of fields ranging from (bio)organic and biophysical chemistry to prebiotic evolution, and from molecular biology to genetic diagnostics and drug development. Many of the applications take advantage of the unique properties of PNA—an uncharged pseudopeptide—that distinguish this DNA mimic from more traditional DNA analogs. Rather than trying to create a comprehensive collection of all published methods and protocols involving PNA—many of which have not yet been validated— I have decided to concentrate on select protocols that are either very well established by several groups around the world, such as PCR-clamping and in situ hybridization, or on new methods that may have broader future impact. Basic methods for PNA oligomer synthesis and analyses have also been included. I am very grateful to those friends and colleagues who have enthusiastically contributed their work, discussions, and writing, and thereby made this book possible. Peter E. Available in PDF, EPUB and Kindle. Book excerpt: In The Aptamer Handbook, leading scientists from academia as well as biotech and pharma companies introduce the revolutionary concept of designing RNA and DNA oligonucleotides with novel functions by in vitro selection. These functions comprise high affinity binding (aptamers), catalytic activity (ribozymes and deoxyribozymes) or combinations of binding and catalytic properties (aptazymes). Basic concepts and technologies describing in detail how these functional oligonucleotides can be identified are presented. Numerous examples demonstrate the versatility of in vitro selected oligonucleotides. Special emphasis has been put on a section that shows the broad applicability of aptamers, e. g. in target validation, for analytics, or as new therapeutics. This first overview in the field is of prime interest for a broad audience of scientists both in academia and in industry who wish to expand their knowledge on the potential of new oligonucleotide functions and their applications.

Available in PDF, EPUB and Kindle. Book excerpt: The essence of combinatorial chemistry or techniques involving "molecular diversity" is to generate enormous populations of molecules and to exploit appropriate screening techniques to isolate active components contained in these libraries. This idea has been the focus of research both in academia and in the pharmaceutical or biotechnology industry. Its developments go hand in hand with an exploding number of potential drug targets emerging from genomics and proteomics research. When the editors of Current Topics in Microbiology and Immunology encouraged us to assemble the present volume on Combinatorial Chemistry in Biology, we immediately felt that this might prove quite beneficial for the audience of this series. The field of combinatorial chemistry extends over a broad range of disciplines, from synthetic organic chemistry to biochemistry, from material sciences to cell biology. Each of these fields may have its own view on this topic, something which is reflected in a growing number of monographs and "special editions" of jour nals devoted to this issue or aspects thereof. The title of the present volume of Springer-Verlag's series suggests that it also has its own special focus. And, generally speaking, this is not wrong: we would even claim the special focus of this volume is on the immunologically relevant aspects of combinatorial chemistry.

Available in PDF, EPUB and Kindle. Book excerpt: Peptide Nucleic Acid (PNA) is an analogue of DNA in which the backbone is a pseudopeptide rather than a sugar. PNA mimics the behaviour of DNA and binds complementary nucleic acid strands. The neutral backbone of PNA results in stronger binding and greater specificity than normally achieved. In addition, the unique chemical, physical and biological properties of PNA have been exploited to produce powerful biomolecular tools, antisense and antigene agents, molecues and biosensors. Important new applications have emerged that could not be performed using oligonucleotides. This book contains state-of-the-art protocols and applications on all aspects of Peptide Nucleic Acids. Concepts are explained clearly and in practical terms and each chapter contains concise background information. Written by leading experts in the field, the book is an invaluable and complete reference work on this novel and exciting area. An essential text for all molecular biologists, biochemists, geneticists, biotechnologists and nucleic acid chemists wishing to keep up with the latest developments.

Available in PDF, EPUB and Kindle. Book excerpt: This book reviews the development, characterization and applications of aptamers in different areas of biotechnology ranging from therapeutics to diagnostics and protein purification. Hailed as chemical antibodies, these single-stranded nucleic acid receptors were predicted to supersede antibodies in traditional assays, such as ELISA, within a short time. While this has yet to happen, readers will find in this book a deep insight into the progress of aptamer technology and a critical discussion about the limitations that need to be overcome in order to find wider acceptance and use outside of the still relatively small aptamer-community. This book covers all aspects of aptamer generation and application for the aptamer-experienced reader and curious novice alike, with the addition of an industry perspective on the future of aptamer-use in biotechnology.
