Book excerpt: Advances in Nanomedicine for the Delivery of Therapeutic Nucleic Acids addresses several issues related to safe and effective delivery of nucleic acids (NAs) using nanoparticles. A further emphasis would be laid on the mechanism of delivery of NAs, the barriers encountered and the strategies adapted to combat them. An exhaustive account of the advantages as well shortcomings of all the delivery vectors being employed in delivery of various NAs will be provided. On final note the regulatory aspects of nanoparticles mediated NA would be discussed, with focus on their clinical relevance. The design and development of nucleic acid-based therapeutics for the treatment of diseases arising from genetic abnormalities has made significant progress over the past few years. NAs have been widely explored for the treatment of cancer and infectious diseases or to block cell proliferation and thereby caused diseases. Advances in synthetic oligonucleotide chemistry resulted in synthesis of NAs that are relatively stable in in vivo environments. However, cellular targeting and intracellular delivery of NAs still remains a challenge. Further development of NA-based therapeutics depends on the progress of safe and effective carriers for systemic administration. Nanomedicine has facilitated availability of vectors with diminished cytotoxicity and enhanced efficacy which are rapidly emerging as systems of choice. These vectors protect NAs from enzymatic degradation by forming condensed complexes along with targeted tissue and cellular delivery. During the past few years, a myriad reports have appeared reporting delivery of NAs mediated by nanoparticles. This book will provide an overview of nanoparticles being employed in the in vitro and in vivo delivery of therapeutically relevant NAs like DNA, siRNA, LNA, PNA, etc. Provides a complete overview of the applicatiosn of nanomedicine in the delivery of nucleic acids, from characterization of nanoparticles, to in vitro and in vivo studies Discusses delivery issues of less well explored nucleic acids, like PNAs, Ribozymes, DNAzymes, etc. Summarizes the current state of research in nucleic acid delivery and underscores the future of nanomedicine in this field

Book excerpt: Nanotechnology and nucleic acid based therapies are two emerging fields in science whose combination has the potential to improve quality of life for patients suffering from various diseases that can so far only be treated in an unsatisfactory way. Nucleic acids offer the potential for highly selective treatment of such diseases or the highly specific modulation of gene expression with RNA interference. A key issue for successful nucleic acid therapies is the availability of a suitable delivery system. Here, the field of nanotechnology offers a multitude of possibilities to develop nanosized delivery vectors tailor-made for various local and systemic approaches. In Nanotechnology for Nucleic Acid Delivery: Methods and Protocols, experts in the field cover the area of nanoparticulate delivery of nucleic acids in terms of biosafety, particle synthesis as well as its application in cell culture. Written in the successful Methods in Molecular BiologyTM series format, chapters include introductions to their respective topics, lists of the necessary materials and reagents, step-by-step, readily reproducible protocols, and notes on troubleshooting and avoiding known pitfalls. Authoritative and easily accessible, Nanotechnology for Nucleic Acid Delivery: Methods and Protocols seeks to serve both professionals and novices with its well-honed methodologies.

Available in PDF, EPUB and Kindle. Book excerpt: This collection of research articles and reviews covers the latest work in the design, delivery, dynamic abilities, and immune stimulation of RNA nanoparticles which have driven the utilization of their immunomodulatory properties. The unknown immune properties of nucleic acid nanoparticles have been a major hurdle in their adaptation until the works herein began assessing their structure-activity relationships. This collection chronologically follows the path of investigating the recognition of design components to implementing them into nucleic acid nanostructures. RNA nanotechnology is an emerging platform for therapeutics with increasing clinical relevance as this approach becomes more widely used and approved for the treatment of various diseases. The latest research aims to take advantage of RNA’s modular nature for the design of nanostructures which can interact with their environments to communicate programmed messages with intracellular pathways. In doing so, nanoparticles can be used to elicit or elude responses by the immune system as desired in conjunction with their therapeutic applications.

Available in PDF, EPUB and Kindle. Book excerpt: Advances in biotechnology have provided scientists with an increasing number of biopharmaceuticals such as novel peptide and protein drugs as well as nucleic acid based drugs for gene therapy. However, successful delivery of these biopharmaceuticals is a major challenge because their molecular properties lead to poor physical and chemical stability in the body and limited membrane permeability. Therefore researchers are developing a range of new delivery technologies and materials to enable these new drugs to be delivered intact to their target sites. Delivery Technologies for Biopharmaceuticals describes strategies to overcome the main barriers for successful delivery of therapeutic peptides, proteins, and nucleic acid-based drugs or vaccines related to the site of administration and the target site. Many of the approaches described are reported in formulations in current clinical trials as well as in marketed products. Contents include: challenges in delivery of biopharmaceuticals novel formulation approaches for peptide and protein injectables non-viral chemical vectors and viral technology for delivery of nucleic acid based drugs immune response, adjuvants and delivery systems for vaccines several examples of delivery systems for different biopharmaceuticals a critical assessment of delivery technologies for biopharmaceuticals Delivery Technologies for Biopharmaceuticals is an essential single-volume introduction to the technologies used by researchers to ensure efficient delivery of this exciting new class of drugs. It will be of value to researchers and students working in drug delivery, formulation, biopharmaceuticals, medicinal chemistry, and new materials development.
