Book excerpt: Surfactants play an essential role in our daily lives. Their form of usage varies from detergents and cleaning agents through disinfectants and solubilisers up to industrial applications such as paints, oil recovery, anti-corrosion protective coatings, etc. A special distinct class of surfactants is represented by a double-chain, double-head form of surfactant molecules, which are called gemini surfactants. Gemini surfactants show physicochemical and aggregation properties which are superior to those of conventional, single-chain surfactants. From the perspective of applications, an important group of gemini surfactants is represented by cationic gemini surfactants. They have found numerous applications in the various fields such as solubilisation, textile coating, organic and polymer synthesis, electrochemistry, paper industry, etc. One of the most developing areas of cationic gemini surfactants application is the field of pharmaceutical applications. Interaction of cationic surfactants with the oppositely charged cell membrane has been known for a long time. Cationic gemini surfactants turned out to be very efficient bactericidal and antimicrobial agents. Moreover, recent development in this field indicates a cancerostatic effect of cationic geminis through a selective interaction of cationic gemini molecules with cancer cells. Another revolutionary field of cationic gemini surfactants application is their interaction with an oppositely charged electrolyte such as DNA. The interaction of DNA with various positively charged systems such as cationic surfactants, polymers and lipids is of great importance with respect to gene transmission through a biological cell membrane to achieve a therapeutic effect in a cell nucleus. The ambition of this monograph is to provide a complex view of synthesis, structure-aggregation, properties-biological activity relationship and recent applications of cationic gemini surfactants in the pharmaceutical field. Individual chapters in the monograph discuss the synthetic preparation of cationic surfactants, the effect of the structure of these compounds on their physical and physicochemical properties, particularly their aggregation properties and associated phenomena. A significant part of this publication is devoted to gemini surfactants, a relatively new class of surfactants whose special and surprising properties increasingly continue to draw the attention of the research community. The final part of the monograph is oriented on the use of cationic surfactants in biomedicine and pharmaceutical applications, where a special emphasis is put on their antimicrobial and antineoplastic activities. Finally, cationic surfactants as potent non-viral gene delivery vehicles are analysed and evaluated. The monograph is intended to serve as a guide for scientists and students in the field of pharmaceutical research and chemistry of colloids and surfactants.

Available in PDF, EPUB and Kindle. Book excerpt: Cationic antimicrobial peptides (CAMPs) are produced by plants, animals and bacteria to protect their host against antagonistic microbes. The antitheses of selective antibiotics, these peptides are drawn by electrostatic and hydrophobic interactions to targets as diverse as the bacterial membrane, nucleic acids and serum proteins. This lack of specificity is their greatest strength, as mutations to single genes rarely lead to bacterial resistance. Resistance may be conferred by large scale alterations in cell envelope composition, which generally reduces bacterial fitness in the absence of peptide. Clinical applications of natural CAMPs are limited, as the peptides are toxic to mammalian cells and rapidly inactivated in vivo by serum albumin and proteases. Faced with these challenges we have prepared a number of CAMP analogues, with the goal of creating lead compounds for further development of antibacterial therapeutics. Much of our work has focused on ultrashort lipopeptides and lipopeptoids, which have properties similar to natural CAMPs and extremely abbreviated sequences. The simple structure of these scaffolds allows rapid creation of CAMP analogues in a brief period of time, allowing us to rapidly explore the structural requirements for CAMP activity. The balance of this work focuses on imparting CAMP-like behaviour to known antibiotics, in order to expand their spectrum of susceptible bacteria and combat the development of drug-resistant bacteria. In particular, the aminoglycosides neomycin and tobramycin have been fused to phenolic disinfectants such as triclosan and biclotymol, in order to improve their diffusion across the bacterial envelope and activity against Gram-negative bacteria.

Book excerpt: This project focused on the development of amphiphiles in order to prevent hospital-acquired infections before they have a chance to enter the host, thereby reducing the need for antibiotics. The antibiotic resistance crisis of the 21st century is a dangerous epidemic with global consequences. Therefore, there is a desperate need for novel approaches in antimicrobial research. This would decrease the overall length of stay in hospitals and costs associated with such a stay. In this study, tetracationic amphiphiles with two 12-carbon chains and an aliphatic linker were studied. The MIC value of each amphiphile and strain pair was determined and compared. Linker length, the distance between head groups attached to identical hydrophobic tails appears to affect antimicrobial activity. Amphiphiles with longer linkers tend to be more potent antibacterial agents than those with shorter linkers. Amphiphiles were also used in combination with antibiotics to determine whether the combinations interacted synergistically to kill bacteria. The most potent amphiphile was 12-B-10-B-12 against all bacteria. This amphiphile was longer in linker length relative to other derivatives, suggesting that there is a relationship between linker length and antimicrobial potency. The development of novel amphiphiles may be one way to reduce the need for antibiotics to treat hospital-acquired infections.

The two most common methods for gene delivery use either synthetic or viral vectors. Viral vectors (infection) are by far the most effective and efficient means of DNA delivery, but their use is tempered by safety and immunogenicity concerns. Consequently, there has been a significant effort to develop and evaluate non- viral vectors, which include cationic amphiphiles and polymers, and more recently anionic amphiphiles. Non-viral vectors have the advantages of ease of production, better stability and low immunogenicity. At the same time, they also have a number of limitations, including low in vitro and in vivo transfection efficiencies, and cytotoxicity in many instances. My research project has been focused on design, development and characterizations of novel amphiphilic lipids for gene delivery. Through rational design and characterization of the amphiphile structures, it not only yielded vectors showing high transfection activities, but also provided information of the structure-activity relationship. These results provide us with a better understanding on the transfection process and future directions to further optimize the amphiphile structures. More specifically, my dissertation research included the following three parts: (i) characterization of novel lipopeptides possessing di- or tri- peptide head groups; (ii) determination of the effect of spacer (between the cationic domain and the hydrophobic domain of charge-reversal amphiphiles) length, rigidity and hydrophilicity on gene delivery; (iii) identification of the cellular uptake pathway and the transfection mechanism of a known enzyme-sensitive charge-reversal amphiphile.
