Available in PDF, EPUB and Kindle. Book excerpt: C.A. VACANTI The loss or failure of an organ or tissue is one of the most frequent, devastating, and costly problems in human health care. Advances in the medical sciences have enabled physicians to restore lost functions in their patients through or gan transplantation, reconstructive surgery with autogenous tissue transfer, or the implantation of alloplastic materials. Although only recently termed "Tissue Engineering" [1], the use of biological andlor synthetic materials in conjunction with cells to create biologic substitutes to serve as functional tis sue replacements has been explored by modern scientists for several decades. The concept of generating functional tissue from an organ biopsy was de scribed very early in the literature: "The Lord God cast a deep sleep on the man, and while he was asleep, he took out one of his ribs and closed up its place with flesh. The Lord God then built up into a woman the rib that he had taken from the man" [2]. Review of history further reveals that, according to legend, the first homotransplantation of an entire limb was performed by Saints Damian and Cosmas, as depicted by the artist Fra Angelico.

Book excerpt: This contributed volume is the first of a series that introduces safe, feasible, and practical decellularization and recellularization techniques for tissue and organ reconstruction. We have put special emphasis on the research areas most likely to develop well-engineered scaffolds for tissue and organ engineering, while presenting easily applicable bench-to-bedside approaches highlighting the latest technical innovations in the field. This book includes both a fundamental discussion for a broad understanding of the basis of tissue repair and substitution, as well as chapters written by world renowned specialists from 20 countries providing deeper discussions and analysis of related sub disciplines. Within these pages, the reader will find state-of-the-art protocols and current clinical challenges in cell and tissue biology, including accurate and comprehensive information on extracellular matrices, natural biomaterials, tissue dynamics, morphogenesis, stem cells, cellular fate progressions, cell and tissue properties for in-vitro and in-vivo applications. This comprehensive and carefully organized treatise provides a clear framework for graduate students and postdoctoral researchers new to the field, but also for researchers and practitioners looking to expand their knowledge on tissue and organ reconstruction.

Book excerpt: This volume provides a state-of-art-report on the new methodologies in tissue engineering and developments in the biomaterials field based on the extracellular matrix-relevant discovery. Extracellular Matrix for Tissue Engineering and Biomaterials opens with an overview of the latest extracellular matrix research and in Part I, focuses on its biology and its role on cell behavior and cell fate relevant for the design of biomimetic surfaces. Part II details issues regarding the strategies currently applied in the research of biologically inspired materials and material systems for the replacement, repair and regeneration of tissues and organs. Part III presents the latest development methods applying knowledge from biology towards nanotechnology, to promote the restoration of the functionality of a living tissue. The book ranges from fundamental biology associated with tissue regeneration for the development of biomimetic approaches to controlling tissue formation, cell function, differentiation and angiogenesis using factors involved in normal tissue development and function. With the breadth and depth of the coverage of this topic, this book will serve as a valuable reference for anyone working in tissue engineering or biomaterials – from scientists, chemists and biologists through physicists, bioengineers and clinicians.

Book excerpt: Soft-tissue reconstruction for a variety of surgical conditions, such as abdominal wall hernia, hiatal hernia, stomal hernia, anal fistula and pelvic floor replacement remains a challenge. There is an insufficient level of high-quality evidence in the literature on the value of bioprosthetics for soft-tissue reconstruction. An expanded knowledge about their clinical efficacy is urgently needed.
